== History == Efficacy was evaluated in TROPION-Breast01 (NCT05104866), a multicenter, open-label, randomized trial. Participants must have experienced disease progression, been deemed unsuitable for further endocrine therapy, and have received one or two lines of prior chemotherapy for unresectable or metastatic disease. Participants were excluded for a history of ILD/pneumonitis requiring steroids, ongoing ILD/pneumonitis, clinically active brain metastases, or clinically significant corneal disease. Participants also were excluded for ECOG performance status >1. Randomization was stratified by previous lines of chemotherapy, prior CDK4/6 inhibitor treatment, and geographical region. A total of 732 participants were randomized (1:1) to datopotamab deruxtecan (n=365) or investigator's choice of chemotherapy (n=367); eribulin (60%), capecitabine (21%), vinorelbine (10%), or gemcitabine (9%). Efficacy was evaluated in a pooled subgroup of 114 participants with locally advanced or metastatic epidermal growth factor receptor-mutated non-small cell lung cancer who had received prior treatment with an epidermal growth factor receptor-directed therapy and platinum-based chemotherapy and received datopotamab deruxtecan at the recommended dose across two clinical trials: TROPION-Lung05 and TROPION-Lung01. == Introduction == Across all living organisms, regulation of gene expression is controlled by interactions between DNA-binding regulatory proteins (transcription factors) and cis-regulatory elements, DNA sequences in or around genes that act as target sites for DNA-binding proteins. By binding to cis-regulatory sequences and to each other, transcription factors fine-tune transcriptional levels by stabilizing/destabilizing binding of RNA polymerase to a gene's promoter. But despite their importance and ubiquity, little is known about where exactly each of these regulatory proteins binds. Literature suggests that nearly 8% of human genes encode transcription factors and the functions and specificities of their interactions remain largely unexplored. We are on the brink of a convergence of high-throughput technologies and genomic theory that is allowing researchers to start mapping these interactions on a genome-wide scale. Only recently has a complete survey of DNA-binding specificities been attempted for a large family of DNA-binding domains. Until the late 1950s, the Precambrian was not believed to have hosted multicellular organisms. However, with radiometric dating techniques, it has been found that fossils initially found in the Ediacara Hills in Southern Australia date back to the late Precambrian. These fossils are body impressions of organisms shaped like disks, fronds and some with ribbon patterns that were most likely tentacles. These are the earliest multicellular organisms in Earth's history, despite the fact that unicellularity had been around for a long time before that. The requirements for multicellularity were embedded in the genes of some of these cells, specifically choanoflagellates. These are thought to be the precursors for all animals. They are highly related to sponges (Porifera), which are the simplest multicellular animals. In order to understand the transition to multicellularity during the Precambrian, it is important to look at the requirements for multicellularity—both biological and environmental.

Hemopressin (Hp) is an alpha hemoglobin fragment with the sequence PVNFKFLSH, originally identified in extracts of rat brain using an enzyme capture technique. It binds cannabinoid receptors, acting as an inverse agonist at CB1 receptors. Longer forms of hemopressin containing 2-3 additional amino acids on the N-terminus have been identified in extracts of mouse brain. These longer hemopressin peptides, named RVD-Hpα and VD-Hpα, bind to CB1 receptors and were originally reported to be agonists. In addition to the Hp peptides from alpha hemoglobin, a related peptide from beta hemoglobin has been found in mouse brain extracts; this peptide, named VD-Hpβ, is also an agonist at CB1 cannabinoid receptors. Hemopressin is not an endogenous peptide but rather an extraction artefact. The only endogenous peptide found endogenously at physiological conditions is RVD-hemopressin (pepcan-12), which has more recently been shown to be a negative allosteric modulator of CB1 receptors and positive allosteric modulator of CB2 receptors. === Rehabilitation and exercise === Cardiac rehabilitation benefits many who have experienced myocardial infarction, even if there has been substantial heart damage and resultant left ventricular failure. It should start soon after discharge from the hospital. The program may include lifestyle advice, exercise, social support, as well as recommendations about driving, flying, sports participation, stress management, and sexual intercourse. Returning to sexual activity after myocardial infarction is a major concern for most patients, and is an important area to be discussed in the provision of holistic care. In the short-term, exercise-based cardiovascular rehabilitation programs may reduce the risk of a myocardial infarction, reduces a large number of hospitalizations from all causes, reduces hospital costs, improves health-related quality of life, and has a small effect on all-cause mortality.
